Confocal Laser Scanning Microscope : CLSM
Principle
Taking advantage of the properties of laser light (short and aligned wavelengths), a laser beam can be focused on a specific position. In addition, the confocal optical system can obtain information on a specific position by collecting beam at the confocal position. Laser beam is scanned in the XY direction to collect information in the XY direction, and then the focal point is moved in the height direction to collect 2-D images at each position. At last ,create a 3-D image by reconstructing theses 2-D images, and obtain roughness information through this process.
